目的观察L型氨基酸转运载体1(LAT1)与其异二聚体CD98hc在胃癌细胞SGC-7901中的表达及LAT1对细胞增殖与迁移能力的影响。方法将胃癌细胞SGC-7901分为1、10、100μmol/L BCH干预和空白对照共4组,RT-PCR、IF与Western blot检测胃癌细胞SGC-7901中LAT1与CD98hc mRNA、蛋白的表达。不同浓度的BCH作用于胃癌细胞SGC-7901 48 h后,新型四唑氮盐(MTS)比色法检测各组细胞生长情况,流式细胞仪检测各组细胞周期分布情况,Transwell细胞迁移小室检测各组细胞迁移能力的变化。结果 LAT1及其异二聚体CD98hc在胃癌细胞SGC-7901中呈高表达。与空白对照组相比,随着BCH浓度的提高,胃癌细胞SGC-7901的增殖能力与迁移能力逐渐降低。细胞周期分析发现,BCH处理的胃癌细胞组S期细胞减少,而G0/G1期细胞增加(P<0.05)。结论 LAT1及其异二聚体CD98hc在SGC-7901细胞中有表达,LAT1可能促进SGC-7901细胞的增殖,加快细胞周期进程,增强SGC-7901细胞的迁移能力。
Objective To investigate the expression of LAT1 and its heterodimer CD98hc in gastric cancer cell line SGC-7901 and the effect of LAT1 on cell proliferation and migration. Methods SGC-7901 cells were divided into 4 groups (1, 10, 100μmol / L BCH intervention) and blank control group. The mRNA and protein expressions of LAT1 and CD98hc in gastric cancer SGC-7901 cells were detected by RT-PCR, IF and Western blot. After different concentrations of BCH were applied to gastric cancer cell SGC-7901 for 48 h, the cell growth of each group was detected by MTS colorimetric assay. The distribution of cell cycle in each group was detected by flow cytometry. Changes of cell migration ability in each group. Results LAT1 and its heterodimer CD98hc were highly expressed in gastric cancer cell line SGC-7901. Compared with the blank control group, the proliferation and migration ability of gastric cancer cell SGC-7901 gradually decreased with the increase of BCH concentration. Cell cycle analysis showed that S phase cells in BCH-treated gastric cancer cells decreased, while cells in G0 / G1 phase increased (P <0.05). Conclusion LAT1 and its heterodimer CD98hc are expressed in SGC-7901 cells. LAT1 may promote the proliferation of SGC-7901 cells, accelerate the cell cycle progression and enhance the migration of SGC-7901 cells.
