采用地高辛原位末端标记法(InSituEndLabeling,ISEL),建立了Apoptosis细胞的原位检测体系。结果显示:小鼠胸腺细胞在体外培养8h时,台盼兰拒染率为94%~95%,地塞米松刺激的胸腺细胞DNA电泳呈典型的“梯形条带(Ladderpatern)”,ISEL标记的细胞阳性率为74.4%,未经地塞米松刺激和未经培养直接标记的胸腺细胞阳性率分别为28.6%和10.2%。
The in situ detection system of Apoptosis cells was established by using InSitu End Labeling (ISEL). The results showed that the trypan blue exclusion rate of mouse thymocytes was 94% ~ 95% when cultured in vitro for 8 h. The DNA ladder of dexamethasone - stimulated thymocytes showed a typical “Ladderpatern” The positive rate of cells was 74.4%. The positive rates of thymocytes without dexamethasone stimulation and direct labeling without culture were 28.6% and 10.2% respectively.
