目的建立功能型饮料中牛磺酸的柱后衍生-高效液相色谱荧光法。方法样品稀释后直接进样,柱后OPA衍生,荧光检测器检测,外标法定量。色谱条件:流动相:乙腈-水(20∶80,V/V);柱后衍生反应温度:55℃;荧光检测器激发波长:330 nm;发射波长:450 nm。结果牛磺酸的浓度为0μg/L~80μg/L时,标准曲线线性关系良好,线性相关系数(r)为0.999 9,检出限为1μg/L,相对标准偏差(RSD)为0.2%~1.0%,加标回收率为97.2%~102.8%,精密度和准确度符合检测要求。结论本方法具有选择性好、精密度高和准确度好等优点,而且由于采用荧光检测器,大大提高了灵敏度,可采取大体积稀释,更易排除其他组分的干扰,更简便,更快捷,适用于日常的检测工作中。
Objective To establish a post-column derivatization-high performance liquid chromatography (HPLC) fluorescence method for taurine in functional beverages. Methods Samples were diluted directly into the sample, post-column derivatization of OPA, fluorescence detector detection, external standard method. Chromatographic conditions: mobile phase: acetonitrile-water (20:80, V / V); post-column derivatization reaction temperature: 55 ℃; fluorescence detector excitation wavelength: 330 nm; emission wavelength: 450 nm. Results When the concentration of taurine was between 0μg / L and 80μg / L, the calibration curve showed a good linearity with a linear correlation coefficient (r) of 0.999 9, a detection limit of 1μg / L and a relative standard deviation (RSD) of 0.2% 1.0%, the recoveries were 97.2% ~ 102.8%, precision and accuracy meet the testing requirements. Conclusion The method has the advantages of good selectivity, high precision and good accuracy, and because of the use of fluorescence detector, greatly increased the sensitivity, can take a large volume dilution, easier to exclude the interference of other components, more simple, faster, Suitable for routine testing work.
