目的 :克隆人 WAF1基因 ,构建成 WAF1 - pc DNA3真核表达质粒。方法 :采用逆转录聚合酶链反应从 Hela细胞中扩增人 WAF1基因 ,并克隆到 pc DNA3真核克隆载体中。结果 :从 Hela细胞扩增的人 WAF1基因克隆至真核克隆表达载体 pc DNA3中 ,经 DNA直接测序 ,获得了 WAF1 -pc DNA3重组质粒。结论 :成功地构建人 WAF1表达质粒 WAF1 - pc DNA3,为进一步研究 WAF1基因表达及生物学效应奠定了基础。
Objective: To clone the human WAF1 gene and construct the eukaryotic expression plasmid of WAF1 - pcDNA3. Methods: The human WAF1 gene was amplified from Hela cells by reverse transcription polymerase chain reaction and cloned into pcDNA3 eukaryotic cloning vector. Results: The human WAF1 gene amplified from Hela cells was cloned into eukaryotic cloning expression vector pcDNA3. After direct DNA sequencing, WAF1-pcDNA3 recombinant plasmid was obtained. Conclusion: The successful construction of the human WAF1 expression plasmid WAF1 - pc DNA3 laid the foundation for further study of WAF1 gene expression and biological effects.
