目的 揭示发育基因Lhx3和Lhx4参与成年动物中枢神经损伤修复的过程。 方法 线栓法制作局灶性脑缺血模型 ,阻断成年大鼠右侧大脑中动脉 ,缺血 2h后再灌注 ,分别于损伤再灌注后 3d、7d、14d和 2 1d取左右侧大脑皮层组织 ,采用RT PCR结合HPLC测定方法 ,相对定量分析上述不同损伤时相同源盒基因Lhx3和Lhx4在损伤侧与对照侧皮层运动神经元mRNA表达量的改变。 结果 正常成年大鼠大脑皮层运动神经元中 ,Lhx3和Lhx4基因均维持一定的表达水平 ,在局灶性脑缺血再灌注 3d、7d、14d后 ,大脑皮层运动神经元损伤侧的Lhx3和Lhx4基因表达均有明显改变 ,但变化规律不同。其中Lhx3在损伤后mRNA表达量较自身对照侧明显降低 ,而Lhx4的mRNA表达量较自身对照侧明显升高 ,该结果与成年大鼠坐骨神经夹伤后 ,脊髓中Lhx3和Lhx4的基因表达变化规律基本一致。 结论 LIM同源盒基因家族中的Lhx3和Lhx4基因不仅在神经元发育过程中起重要调控作用 ,而且可能参与成年动物中枢神经损伤的修复过程。
Objective To reveal the involvement of developmental genes Lhx3 and Lhx4 in the repair of central nervous system injury in adult animals. Methods The focal cerebral ischemia model was established by the method of thread occlusion. The right middle cerebral artery was occluded in adult rats. After 2h of ischemia, reperfusion was performed. The left and right cerebral cortex were harvested at 3d, 7d, 14d and 21d after reperfusion RT-PCR combined with HPLC method was used to quantitatively analyze the changes of mRNA expression of Lhx3 and Lhx4 in the injured and contralateral cortical neurons in the same injury-induced injury. Results The expression of Lhx3 and Lhx4 genes in normal adult rat cortical motor neurons maintained a certain level. After focal cerebral ischemia and reperfusion for 3d, 7d and 14d, Lhx3 and Lhx4 Gene expression were significantly changed, but the variation is different. The mRNA expression of Lhx3 in the injured group was significantly lower than that of the control group, while the expression of Lhx4 mRNA was significantly higher than that in the control group. The results showed that the gene expression of Lhx3 and Lhx4 in the spinal cord changed with the sciatic nerve injury Basically the same. Conclusions Lhx3 and Lhx4 genes in the LIM homeobox gene family not only play an important regulatory role in the development of neurons, but also may participate in the repair of central nervous system injury in adult animals.
