目的:比较细胞增殖相关基因n Twist1、SIRT1、FGF2、TGF-β3在胎盘、脐带和乳牙3种间充质干细胞中的表达差异,分析其对3种间充质干细胞的增殖调控作用。n 方法:通过显微镜对不同传代次数的3种间充质干细胞的形态结构进行比较;经过体外传代培养,利用荧光定量PCR测定细胞增殖相关基因n Twist1、SIRT1、FGF2、TGF-β3的表达水平;采用MTT法检测并比较分析3种间充质干细胞的增殖及促增殖能力。n 结果:Twist1、TGF-β3基因在胎盘间充质干细胞中表达水平最高,n FGF2基因的表达水平最低,n SIRT1基因在脐带间充质干细胞中的表达水平较高。随着传代培养进行,4种基因表达水平在不同代次的间充质干细胞中的表达水平各不相同。n Twist1、SIRT1、TGF-β3表达上调时3种间充质干细胞增殖能力增强,n FGF2表达上调时3种间充质干细胞增殖能力减弱。n 结论:Twist1、SIRT1、FGF2、TGF-β3在3种间充质干细胞中的表达水平存在一定差异;n Twist1、SIRT1、TGF-β3对3种间充质干细胞的增殖表现为正调控作用;而n FGF2基因则相反表现为负调控作用。n “,”Objective:To compare the mRNA level of cell proliferation-related genes n Twist1, SIRT1, FGF2 and n TGF-β3 in placenta mesenchymals stem cells (PA-MSCs), umbilical cord mensenchymals (UC-MSCs) and dental pulp mesenchymals stem cells (DP-MSCs).n Methods:The morphology of various passages of PA-MSCs, UC-MSCs and DP-MSCs were observed by microscopy. Proliferation and promoting ability of the three cell lines were detected with the MTT method. Real-time PCR (RT-PCR) was used to determine the mRNA levels of n Twist1, SIRT1, FGF2, TGF-β3.Results:The morphology of UC-MSCs and DP-MSCs was different from that of PA-MSCs. Proliferation ability and promoting ability of the PA-MSCs was superior to that of UC-MSCs and DP-MSCs. In PA-MSCs, expression level of n Twist1 and n TGF-β3 was the highest and n FGF2 was the lowest. n SIRT1 was highly expressed in UC-MSCs. With the cell subcultured, different expression levels of n Twist1, SIRT1, FGF2, TGF-β3 was observed in PA-MSCs, UC-MSCs and DP-MSCs.n Conclusion:Up-regulated expression of the n Twist1, SIRT1 and n TGF-β3 genes can promote proliferation of PA-MSCs, UC-MSCs and DP-MSCs, whilst n TGF-β3 may inhibit these. The regulatory effect of n Twist1, SIRT1, FGF2 and n TGF-β3 genes on PA-MSCs, UC-MSCs and DP-MSCs are different.n
