目的建立反相高效液相色谱法同时测定九味羌活丸中升麻素苷、升麻素、黄芩苷的含量的测定方法。方法采用Phenomenex Luna C18色谱柱(250mm×4.6mm,2.5μm),流动相为乙腈∶0.05%磷酸水溶液=25∶75,流速为1.0ml/min,柱温为室温,检测波长为256nm。结果升麻素苷的线性回归方程为:Y=4.216×10~4X-8.362×102(r=0.9998),在0.691~6.91mg/L范围内呈良好的线性关系;升麻素线性回归方程为:Y=6.529×10~4-1.201×103(r=0.9997),在0.326~3.26mg/L范围内呈良好的线性关系;黄芩苷的线性回归方程为:Y=2.703×10~4-7.182×10~4(r=0.9996),在21.1~211mg/L范围内呈良好的线性关系。结论反相高效液相色谱法测定九味羌活丸中三种有效成分的含量,操作方法简单,精密度高,重现性好,可以作为九味羌活丸的质量控制方法。
Objective To establish a method for the simultaneous determination of cinnarinol, cimicifuguin and baicalin in Jiuwei Notopterygium by reversed-phase high performance liquid chromatography. Methods Phenomenex Luna C18 column (250mm × 4.6mm, 2.5μm) was used. The mobile phase consisted of acetonitrile: 0.05% phosphoric acid aqueous solution = 25:75, the flow rate was 1.0ml / min, the column temperature was room temperature and the detection wavelength was 256nm. Results The linear regression equation of cilengine was: Y = 4.216 × 10 ~ 4X-8.362 × 102 (r = 0.9998), which showed a good linear relationship in the range of 0.691 ~ 6.91mg / L; the linear regression equation : Y = 6.529 × 10 ~ 4-1.201 × 103 (r = 0.9997). The linear regression equation of baicalin was Y = 2.703 × 10 ~ 4-7.182 × 10 ~ 4 (r = 0.9996), in the range of 21.1 ~ 211mg / L showed a good linear relationship. Conclusion The RP-HPLC method for the determination of the content of the three effective components in Jiuwei Notopterygium pills is simple, accurate and reproducible. It can be used as quality control method for Jiuwei Notopterygium.
