用分光光度计终点测定法和酶标仪动力学测定法对3个抗性水平不同的棉蚜品系和1个敏感品系的羧酸酯酶进行了研究。棉蚜的不同抗性品系羧酸酯酶对α-乙酸萘酯(α-NA)和β-乙酸萘酯(β-NA)的水解活性具有极显著的相关性(r=0.95,n=400)。两种测定法均显示出R1、R2和R3品系的羧酸酯酶比活力明显高于S品系。以α-NA为底物时,用酶标仪动力学测定法研究表明,S、R1、R2和R3棉蚜品系羧酸酯酶活性分别为38、85、198和762mOD·min~(-1)·aphid~(-1);与4个品系的抗性程度比较,酶动力学方法的测定结果更可靠。
Carboxylesterase activities of three susceptible cotton aphid strains and one susceptible strain were studied by spectrophotometer endpoint assay and microplate reader kinetic assay. The carboxylesterase resistance of different resistant strains of cotton aphid had a very significant correlation (r = 0.95, n = 400) for α-NA and β-NA hydrolysis activity ). Both assays showed that the carboxylesterase activities of R1, R2 and R3 lines were significantly higher than those of S lines. When α-NA was used as a substrate, the carboxylesterase activity of the cotton aphid strains S, R1, R2 and R3 was 38, 85, 198 and 762 mOD · min ~ (-1) ) · Aphid ~ (-1). Compared with the resistance of the four strains, the results of enzyme kinetic method are more reliable.
