目的以主要表面糖蛋白上游保守区UCS基因对中国耶氏肺孢子菌进行基因多态性分析。方法用巢式PCR扩增127例耶氏肺孢子菌感染患者呼吸道标本中主要表面糖蛋白上游保守区UCS基因,通过DNA直接测序获得该基因的序列,并随机选取部分标本经过克隆测序,分析其UCS串联重复区序列的特征,并分析不同患者人群中基因多态性分布。结果共获得97例(占76.4%)患者标本的UCS基因序列,依据串联重复单元的数目、序列不同得到2~5个重复的7种基因型,其中基因型1,2,3(占68.04%)最为常见,其次为基因型1,2,3,3(占21.65%)。在6组患者中,基因型1,2,3亦最为常见。结论本研究获得了我国耶氏肺孢子菌UCS基因多态性的基础资料,为进一步分析耶氏肺孢子菌的遗传学、流行病学提供了依据。
OBJECTIVE: To analyze the polymorphism of Pneumocystis yerstenii from UCS gene upstream of the major surface glycoprotein. Methods UCS gene was amplified by nested PCR in 127 cases of respiratory tract samples of respiratory tract infected by Pneumocystis pneumoniae. The sequence of this gene was obtained by direct sequencing of DNA, and some of the samples were randomly selected for cloning and sequencing UCS characteristics of tandem repeat regions and to analyze the distribution of gene polymorphisms in different patient populations. Results A total of 97 cases (76.4%) of UCS gene sequences were obtained. According to the number and sequence of tandem repeat units, there were 7 genotypes with 2 to 5 repeats, of which genotypes 1, 2 and 3 (68.04% ) Were the most common, followed by genotype 1,2,3,3 (21.65%). Of the 6 groups of patients, genotypes 1, 2 and 3 were the most common. Conclusions This study obtained the basic data of UCS gene polymorphism of Pneumocystis yersuioi in China, which provided the basis for further analysis of the genetics and epidemiology of Pneumocystis sp.
