目的:以纤维蛋白胶为载体复合胰岛素样生长因子-Ⅰ,研究该复合物对大鼠成骨细胞形态、粘附、增殖和分化的影响。方法:制备纤维蛋白胶-IGF-Ⅰ复合物,按实验分组:纤维蛋白胶-IGF-Ⅰ复合物组;纤维蛋白胶组;IGF-Ⅰ组;空白对照组,与SD乳鼠颅顶骨成骨细胞共同体外培养,采用噻唑蓝比色法、碱性磷酸酶活性及骨钙素分泌检测法以比较各组成骨细胞增殖和分化的情况。结果:第1天,四组间成骨细胞增殖及碱性磷酸酶活性差异无统计学意义,纤维蛋白胶-IGF-Ⅰ复合物组骨钙素量(OC值)与IGF-Ⅰ组差异无统计学意义,但高于纤维蛋白胶组和空白对照组,差异有统计学意义(P<0.05);第3d、5d、7d,纤维蛋白胶-IGF-Ⅰ复合物组成骨细胞增殖活力、碱性磷酸酶活性、骨钙素量均高于其余各组,差异有统计学意义(P<0.05)。结论:纤维蛋白胶有强大的粘附作用,能复合生长因子IGF-Ⅰ,并能通过逐步降解缓慢释放生长因子,促进成骨细胞的增殖与分化。
OBJECTIVE: To investigate the effects of the complex on the morphology, adhesion, proliferation and differentiation of rat osteoblasts by using insulin-like growth factor-I as fibrin glue. METHODS: Fibrin glue-IGF-Ⅰ complex was prepared and divided into groups according to the experiment: fibrin glue-IGF-Ⅰ complex group, fibrin glue group, IGF-Ⅰ group, blank control group, The cells were cultured in vitro, and the proliferation and differentiation of osteoblasts were compared using thiazolyl blue colorimetric assay, alkaline phosphatase activity and osteocalcin secretion assay. Results: There was no significant difference in osteoblast proliferation and alkaline phosphatase activity between the four groups on the first day. There was no significant difference between osteocalcin (IGF-Ⅰ) group and fibrin glue-IGF-Ⅰ complex group (P <0.05). On the 3rd, 5th and 7th day, the fibrin glue-IGF-Ⅰ complex formed the proliferation activity of the osteoblasts, and the alkali The activity of phosphatase and osteocalcin were higher than the other groups, the difference was statistically significant (P <0.05). CONCLUSION: Fibrin glue has a strong adhesive effect and can complex growth factor IGF-Ⅰ. It can promote the proliferation and differentiation of osteoblasts through gradual degradation and slow release of growth factors.
