目的考察黄芩(SBE)提取物对D-半乳糖和亚硝酸钠合并诱导的阿尔茨海默氏病(AD)模型小鼠抗氧化能力的保护作用。方法腹腔注射D-半乳糖150 mg·kg-1和亚硝酸钠100 mg·kg-1,连续60 d,制备小鼠AD模型。并在小鼠造模当天开始,灌胃低、中、高3个剂量(125,250,500 mg·kg-1·d-1)SBE,同样连续60 d。在造模、给药第60天,将小鼠摘眼球取血1.5 m L。用超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、丙二醛(MDA)试剂盒测定小鼠血清中SOD、GSH-Px和CAT活力、以及MDA水平。结果在AD模型组中,小鼠血清中SOD、GSH-Px和CAT的活力显著降低而MDA的含量升高。S高剂量BE组可显著性增强AD模型小鼠血清中SOD的活力;中、高2个剂量SBE组可剂量依赖性的显著增强AD模型小鼠血清中GSH-Px和CAT的活力,并显著性降低MDA的含量。结论 SBE可以剂量依赖性的提高AD模型小鼠体内的抗氧化能力。
Objective To investigate the protective effect of Scutellaria baicalensis (SBE) extract on the antioxidant capacity in Alzheimer’s disease (AD) model mice induced by D-galactose and sodium nitrite. Methods The mouse model of AD was prepared by intraperitoneal injection of 150 mg · kg -1 D-galactose and 100 mg · kg -1 sodium nitrite for 60 days. At the beginning of the day of modeling, SBE at three levels (125, 250 and 500 mg · kg-1 · d-1) were given intragastrically for the same duration of 60 days. On the 60th day of administration, mice were ejaculated and blood was drawn for 1.5 m L at the time of modeling. The levels of SOD, GSH-Px in serum of mice were determined by the method of SOD, GSH-Px, CAT and MDA. And CAT activity, as well as MDA levels. Results In the AD model group, the activities of SOD, GSH-Px and CAT in the serum of mice were significantly decreased and MDA content was increased. S high dose BE group can significantly enhance the activity of SOD in AD model mice serum; middle and high doses of SBE group dose-dependently significantly increased GSH-Px and CAT activity in AD model mice serum, and significantly Sex reduces MDA content. Conclusion SBE can increase the antioxidant capacity of AD mice in a dose-dependent manner.
