目的建立HPLC-ELSD法测定更年调和颗粒中黄芪甲苷的含量。方法色谱柱为Thermo Syncronis C18柱(250 mm×4.6 mm,5μm);流动相为乙腈-水(34∶66);流速:1.0 mL·min-1;柱温:30℃。蒸发光散射检测器(ELSD)参数:漂移管温度85℃,氮气体积流量1.8 mL·min-1。结果黄芪甲苷在0.812~8.12μg范围内具有良好的线性关系,加样回收率为98.07%(RSD为1.21%,n=6)。结论本法简单易行,重复性符合要求,测定结果准确,可作为更年调和颗粒中黄芪甲苷的含量测定方法。
OBJECTIVE To establish HPLC-ELSD method for the determination of Astragaloside Ⅳ in Granule for the Year of the Masses. Methods The column was Thermo Syncronis C18 (250 mm × 4.6 mm, 5 μm). The mobile phase was acetonitrile-water (34:66). The flow rate was 1.0 mL · min-1. The column temperature was 30 ℃. Evaporative Light Scattering Detector (ELSD) Parameters: drift tube temperature 85 ° C, nitrogen volume flow 1.8 mL · min-1. Results Astragaloside had a good linear relationship in the range of 0.812 ~ 8.12 μg with the recovery of 98.07% (RSD 1.21%, n = 6). Conclusion This method is simple, reproducible, and accurate. It can be used as a method for determining the content of Astragaloside IV in Granules of Greater Year.
