目的:观察opticin在1型、2型糖尿病小鼠及正常小鼠玻璃体视网膜mRNA和蛋白表达的变化。方法:50只雄性小鼠随机分为柠檬酸钠缓冲液对照组(A)10只,链脲佐菌素诱导1型糖尿病组(B)10只,2型糖尿病组30只,并将其随机分为1,2,3月组(C、D、E),各10只。于1,2,3月后处死2型糖尿病组小鼠,3月后处死1型糖尿病组小鼠和柠檬酸钠缓冲液对照组,取玻璃体视网膜组织,保存于液氮中。实时聚合酶链反应检测玻璃体视网膜中opticin mRNA表达,Western-blot检测opticin蛋白表达。结果:1型糖尿病组玻璃体视网膜的opticinmRNA表达量是正常组玻璃体视网的68%,P<0.01;2型糖尿病1,2,3月组玻璃体视网膜的opticin mRNA表达量分别是正常组玻璃体视网的37%,36%,27%,P<0.01;但1,2,3月组之间的差异无统计学意义,P>0.05;1型、2型糖尿病组玻璃体视网膜的opticin蛋白表达量较正常小鼠玻璃体视网膜显著下降,P<0.01。2型糖尿病的2月、3月组小鼠玻璃体视网膜mRNA和蛋白表达均低于1月组小鼠。结论:1型、2型糖尿病小鼠玻璃体视网膜中opticin的mRNA和蛋白表达量较正常小鼠显著减少,2型糖尿病组小鼠玻璃体视网膜mRNA和opticin蛋白表达随时间延长逐渐降低。说明opticin在糖尿病视网膜病变的发生机制中可能起着重要作用。
Objective: To observe the changes of vitreoretinal mRNA and protein expression in optic nerve of type 1 and type 2 diabetic mice and normal mice. Methods: Fifty male mice were randomly divided into 10 groups: control group (A), sodium citrate buffer group (10), streptozotocin induced type 1 diabetes mellitus group (B), type 2 diabetic group (30) Divided into 1, 2, 3 months group (C, D, E), each 10. Type 1 diabetic mice were sacrificed in January, February and March, and mice in type 1 diabetic group and sodium citrate buffer group were sacrificed after 3 months. Vitreoretinal tissue was taken and stored in liquid nitrogen. The expression of opticin mRNA in vitreoretinal was detected by real-time polymerase chain reaction. The protein expression of opticin was detected by Western-blot. Results: The expression level of opticinmRNA in vitreoretinal of type 1 diabetes mellitus group was 68% of that in normal group, P <0.01. The expressions of opticin mRNA in vitreoretinal group in type 1 diabetes mellitus were normal vitreoretinal Compared with 37%, 36%, 27%, P <0.01, but there was no significant difference between the 1, 2 and 3 months group, P> 0.05; the protein expression of vitreoretinal in type 1 and type 2 diabetic patients The vitreous retina of normal mice decreased significantly, P <0.01. The expression of vitreoretinal mRNA and protein of the mice in 2 month and 3 month group were lower than those in the 1 month group. CONCLUSION: The expression of opticin mRNA and protein in vitreoretinal retina of mice with type 1 and type 2 diabetes is significantly reduced compared with that of normal mice. The expression of vitreoretinal mRNA and optic protein in mice with type 2 diabetes mellitus gradually decreases with time. It shows that opticin may play an important role in the pathogenesis of diabetic retinopathy.
