目的对银屑病自身抗原--角蛋白17的HLA-DR限制性T细胞表位区进行确定. 方法借助相关软件和互联网服务器进行角蛋白17的辅助性T细胞抗原表位区预测;从银屑病患者皮损组织中扩增出人角蛋白17全长基因;根据预测结果,融合表达、纯化相应的表位区;分离纯化外周血T淋巴细胞,将各表位区肽段分别与正常人和银屑病患者的T淋巴细胞体外作用,检测T细胞增殖程度和培养上清中IFN-γ水平变化,从而筛选出特异性强反应性表位区. 结果角蛋白17的 65～120肽段、305～374肽段能够显著促进银屑病患者T淋巴细胞增殖和IFN-γ分泌,与正常对照组比较差异有非常显著性(均为P
