目的 以血管内皮生长因子 (vascular endothelial growth factor,VEGF)抗体进行抗血管生成治疗胶质瘤的研究。方法 应用 VEGF抗体分别作用于 C6胶质瘤细胞系的体外培养细胞和动物模型 ,计算细胞存活率及抑瘤率 ,ABC免疫组化技术检测体内生长胶质瘤中微血管密度。结果 VEGF抗体不抑制体外培养细胞生长 ,细胞存活率为 10 0 %。对体内生长的胶质瘤则有显著抑制作用且呈剂量依赖关系 ,抑瘤率可达 82 .8% (P<0 .0 1)。经VEGF抗体治疗的胶质瘤中微血管密度较对照组降低 (P<0 .0 1)。结论 VEGF抗体能抑制胶质瘤血管生成 ,在胶质瘤临床治疗中可能有应用价值。
Objective To study antiangiogenic therapy for gliomas with vascular endothelial growth factor (VEGF) antibody. Methods VEGF antibody was applied to cultured cells and animal models of C6 glioma cell lines in vitro. Cell viability and tumor inhibition rate were calculated. ABC immunohistochemistry was used to detect microvessel density in growing gliomas. Results VEGF antibody did not inhibit cell growth in vitro and the cell survival rate was 100%. The growth of gliomas in vivo was significantly inhibited in a dose-dependent manner, and the tumor inhibition rate was 82.8% (P < 0.01). The microvessel density in glioma treated with VEGF antibody was lower than that in the control group (P < 0.01). Conclusion VEGF antibody can inhibit the angiogenesis of glioma, which may have application value in the clinical treatment of glioma.
