目的研究紫杉醇对体外培养的高转移性肝癌细胞株MHCC97-H中ezrin的表达的影响。方法将体外培养的MHCC97-H分为4组,分别与不同浓度的紫杉醇共培养,然后用免疫组化、Western blot法、RT-PCR检测各组中ezrin的表达水平,Transwell穿膜实验检测肝癌细胞侵袭能力的变化。结果正常培养的MHCC97-H中有ezrin的高表达,紫杉醇可以抑制MHCC97-H中的ezrin表达,经紫杉醇处理的肝癌细胞穿透人工膜的能力下降。结论紫杉醇可以抑制体外培养的MHCC97-H中ezrin的表达,从而抑制肝癌细胞的侵袭能力。
Objective To study the effect of paclitaxel on the expression of ezrin in MHCC97-H cell line with high metastatic potential. Methods MHCC97-H cultured in vitro were divided into 4 groups and co-cultured with different concentrations of paclitaxel. The expression of ezrin in each group was detected by immunohistochemistry, Western blot and RT-PCR. Changes in cell invasiveness. Results High expression of ezrin was found in normal cultured MHCC97-H. Paclitaxel could inhibit ezrin expression in MHCC97-H, and the ability of paclitaxel-treated hepatoma cells to penetrate artificial membrane decreased. Conclusion Paclitaxel can inhibit the expression of ezrin in MHCC97-H cultured in vitro and thus inhibit the invasion ability of hepatoma cells.
