目的 :建立测定血浆中丙戊酸血药浓度的高效液相色谱法。方法 :血浆用正己烷提取 ,经α-溴苯乙酮衍生后浓集进样 ,采用 Alltech C1 8柱 (15 0 mm× 4.6 m m,5︼m ) ,以甲醇∶水 (80∶ 2 0 )为流动相 ,环己烷羧酸为内标物 ,在波长 2 48nm处测定。结果 :丙戊酸的线性范围为 11.31~ 181.0 0︼g/ m l,平均回收率大于 97% ,日内和日间 RSD<5 %。结论 :本法快速、简便、准确 ,适用于丙戊酸血药浓度测定
Objective: To establish a HPLC method for the determination of plasma valproate in plasma. Methods: Plasma samples were extracted with n-hexane and concentrated with α-bromoacetophenone. Alltech C1 8 columns (150 mm × 4.6 mm, 5 μm) As mobile phase, cyclohexanecarboxylic acid as an internal standard, measured at a wavelength of 482nm. Results: The linear range of valproic acid was 11.31 ~ 181.0 0︼g / m l. The average recovery was more than 97%. The intra-day and inter-day RSD was less than 5%. Conclusion: This method is rapid, simple and accurate and is suitable for the determination of plasma concentration of valproic acid
