目的:研究蒙药忠伦阿汤对胶原诱导性(CIA)关节炎大鼠血清及关节中OPG/RANKL的影响。方法:将CIA大鼠模型分为正常组、模型组、蒙药忠伦阿汤高、中、低剂量组及雷公藤多苷片组。实验过程中定期观察大鼠的一般活动情况。ELISA法检测大鼠血清中抗CII抗体及OPG和RANKL的含量。大鼠踝关节病理切片HE染色光镜观察滑膜和软骨组织病理学形态结构的变化,免疫组化SP法检测大鼠踝关节中OPG及RANKL的表达情况。结果:造模大鼠血清中抗CII抗体含量均高于正常组;正常组大鼠血清及踝关节中OPG呈高表达而RANKL呈低表达,模型组大鼠OPG呈低表达而RANKL呈高表达,二者有显著差异(P<0.05),各治疗组药物均可下调RANKL的表达并升高OPG的表达,使OPG/RANKL比值增大。结论:蒙药忠伦阿汤能够一定程度增加血清及关节中OPG的表达而减少RANKL的表达,使OPG/RANKL的比值增大。蒙药忠伦阿汤可能是通过主要抑制RANKL的表达并辅助增强OPG的表达,使OPG/RANKL比例增大,OPG能够竞争性结合RANKL而阻断RANK与RANKL的结合,来实现缓解或延缓类风湿关节炎的病情发展。
Objective: To investigate the effect of Mongolian medicine Zhong Lun A Tang on OPG / RANKL in serum and joint of collagen-induced arthritis (CIA) rats. Methods: CIA rats were divided into normal group, model group, Mongolian Zhonglun A Tang high, medium and low dose groups and Tripterygium tablets group. During the experiment regularly observe the general activity of rats. Serum anti-CII antibody and OPG and RANKL contents were measured by ELISA. Pathological changes of synovium and cartilage histopathology were observed with HE staining and pathological sections of rats ankle. The expression of OPG and RANKL in ankle joint of rats were detected by immunohistochemical SP method. Results: The levels of anti-CII antibody in the serum of model rats were higher than that of normal rats. In normal rats, OPG and RANKL were low in serum and ankle joints, but low in OPG and high in RANKL (P <0.05). The drugs of each treatment group could down-regulate the expression of RANKL and increase the expression of OPG, so that the ratio of OPG / RANKL increased. Conclusion: Mongolian medicine Zhong Lun A decoction can increase the expression of OPG in serum and joint to reduce the expression of RANKL, and increase the ratio of OPG / RANKL. Mongolia Mongolian Zhonglun Decoction may be through the main inhibition of RANKL expression and help enhance the expression of OPG, OPG / RANKL ratio increased, OPG can compete with RANKL and block the combination of RANK and RANKL, to achieve or delay rheumatoid Arthritis progression.
