目的观察16Hz、130dB次声对小鼠记忆功能和脑超微结构的影响及不同剂量姜黄素的治疗作用。方法Morris水迷宫成绩相近的BALB/C小鼠接受16Hz、130dB的次声作用,2h/d,同时给予不同剂量的姜黄素,14d后再次评定水迷宫成绩,并取大脑额叶皮层做透射电镜,观察小鼠脑超微结构的变化。结果单纯次声暴露后,小鼠记忆功能下降(和空白组相比,P<0.05),神经细胞缺血性改变,胞浆内脂褐素增加,髓鞘变性,毛细血管水肿;姜黄素各组记忆功能改善(与单纯次声组比较,P<0.05),神经细胞轻度或没有缺血性改变,脂褐素少或没有,髓鞘变性及毛细血管水肿无明显改善。结论16Hz、130dB次声可引发小鼠脑皮质的脂质过氧化,使记忆功能受损。姜黄素可明显减轻次声引发的这些损害,机制可能与其抗氧化作用有关。其治疗作用有一定的部位选择性,主要集中在神经细胞内。
Objective To observe the effect of 16Hz and 130dB infrasound on memory function and brain ultrastructure of mice and the therapeutic effect of different doses of curcumin. Methods The BALB/C mice with similar performance in the Morris water maze received 16Hz, 130dB infrasound, 2h/d, and different doses of curcumin. After 14 days, the water maze performance was assessed again. The frontal cortex was taken as the transmission electron microscope. Observe the changes of mouse brain ultrastructure. Results After simple infrasound exposure, the memory function of the mice was decreased (P<0.05 compared with the blank group), the ischemic changes of the neurons, the increase of cytosolic lipofuscin, the demyelination of the myelin sheath, and the capillary edema; each of the curcumin Improvement in group memory (compared with simple infrasound group, P<0.05), slight or no ischemic changes in neurons, little or no lipofuscin, no significant improvement in myelin degeneration and capillary edema. Conclusion The 16Hz and 130dB infrasound can induce lipid peroxidation in mouse cerebral cortex and impair the memory function. Curcumin can significantly reduce the damage caused by infrasound, and the mechanism may be related to its anti-oxidation effect. The therapeutic effect has a certain site selectivity, mainly concentrated in nerve cells.
