SMMC-7721人体肝癌细胞系是第二军医大学于1977年8月开始培养建立,材料取自二军大第一附属医院的一例肝癌手术标本。至今在体外连续传代培养已46个月,液氮低温冻存37个月。该细胞系的生长较为迅速而稳定,每隔7~10天可传代一次,细胞形态以上皮型为主,其亚微结构符合肝癌细胞的特征。本细胞系在第15代时甲胎蛋白和白蛋白的间接免疫荧光反应阳性,同时用放射免疫法测得培养液中的甲胎蛋白为31毫克/毫升。本系细胞培养220天时,染色体数目为44~107个,主流范围在54258个,占总数的70%,染色体核型分析,增加的大部属A、B、C,E和F群,其中C、E群的增加和G群的减少较突出。本系细胞乳酸脱氢酶同功酶谱符合肝癌细胞的一般
The SMMC-7721 human hepatoma cell line was established in the Second Military Medical University in August 1977 and the material was taken from a case of liver cancer surgery at the First Affiliated Hospital of the Second Military University. Forty-six months after continuous serial subculture in vitro, liquid nitrogen was cryopreserved for 37 months. The growth of this cell line is relatively rapid and stable. It can be passaged once every 7 to 10 days. The cell morphology is predominantly skin type, and its sub-microstructure conforms to the characteristics of hepatoma cells. The indirect immunofluorescence reaction of alpha-fetoprotein and albumin was positive in the 15th generation of the cell line, and the alpha-fetoprotein in the culture fluid was 31 mg/ml as determined by radioimmunoassay. At 220 days of culture, the number of chromosomes was 44 to 107, and the main range was 54258, accounting for 70% of the total number. The karyotype analysis showed that most of the genes were added to the groups A, B, C, E, and F, of which C, The increase of E group and the decrease of G group are more prominent. The lactic acid dehydrogenase isoenzyme spectrum of this lineage is consistent with that of liver cancer cells.
