目的 研究视网膜母细胞瘤 ( retinoblastoma,RB)移植瘤中的细胞增殖活性与其发生发展及浸润生长的相关性。方法 对 2 0例裸小鼠玻璃体腔 RB移植瘤进行细胞增殖因子 ki-67蛋白表达的免疫组织化学染色 ,以正常视网膜组织标本作为正常对照。比较分析 RB移植瘤眼内生长期 ( ~ 级 )及蔓延扩展浸润期 ( ~ 级 ) ki-67蛋白表达的情况。结果 正常视网膜组织中 ki-67蛋白表达阴性。 2 0例 RB移植瘤细胞的细胞核呈棕黄色 ki-67蛋白阳性表达。在 RB移植瘤生长的不同阶段从 ~ 级 ,ki-67蛋白阳性表达细胞数逐渐增多。眼内生长期 ( ~ 级 )与眼内蔓延眼外转移期 ( ~ 级 ) ki-67蛋白阳性表达细胞数有明显统计学差异 ( P<0 .0 5)。结论 ki-67蛋白表达与 RB移植瘤本身生物学行为有关。RB移植瘤的蔓延浸润能力与其细胞的增殖活性有关 :增殖活性越高 ,其浸润潜能越大
Objective To study the relationship between cell proliferative activity and its development and invasion and growth in retinoblastoma (RB) transplanted tumor. Methods Immunohistochemical staining of cell proliferation factor ki-67 protein was performed on vitreous cavity RB xenografts in 20 nude mice. Normal retinal tissue specimens were used as normal control. The expression of ki-67 protein in the growth phase (~ grade) and spreading extended infiltration stage (~ grade) of RB xenografts was compared and analyzed. Results The normal retinal tissue ki-67 protein negative expression. The nuclei of 20 xenografted RB tumor cells were positive for ki-67 protein. In the different stages of the growth of RB xenografts, the number of positive expression of ki-67 protein gradually increased from ~ level. The number of positive expression of ki-67 protein in the intraocular (~ grade) and intraocular (i) grade eyes had a statistically significant difference (P <0.05). Conclusions The expression of ki-67 protein is related to the biological behavior of RB xenografts. The invasive and infiltrating capacity of RB xenografts is related to the proliferation activity of its cells: the higher the proliferative activity, the greater its infiltration potential
