目的探讨FAT10过表达对肝癌细胞HepG2侵袭及迁移的影响。方法采用脂质体转染法建立FAT10基因过表达的HepG2细胞株;采用蛋白质印迹法检测转染重组表达载体pcDNA3-FAT10后,HepG2细胞中FAT10蛋白的表达水平。体外侵袭实验及划痕实验检测FAT10基因过表达对HepG2细胞迁移及侵袭能力的改变,并采用芯片检测FAT10下游与细胞侵袭转移相关基因的差异表达。结果蛋白质印迹法检测结果显示,转染重组表达载体pcDNA3.1-FAT10的HepG2细胞中FAT10蛋白均过表达;FAT10过表达能明显提高HepG2细胞的侵袭及迁移能力。结论 FAT10基因在HepG2细胞中过表达能提高肝癌的侵袭及迁移能力,并促进肝癌的发生及发展。
Objective To investigate the effect of FAT10 over-expression on the invasion and migration of HepG2 cells. Methods HepG2 cells with FAT10 gene overexpression were established by lipofection method. The expression of FAT10 protein in HepG2 cells was detected by Western blotting after transfected with recombinant pcDNA3-FAT10. In vitro invasion assay and scratch assay were used to detect the changes of FAT10 gene overexpression on the migration and invasion ability of HepG2 cells. The differentially expressed genes related to the invasion and metastasis of FAT10 in the downstream of FAT10 were detected by microarray. Results The results of Western blotting showed that FAT10 protein was over-expressed in HepG2 cells transfected with pcDNA3.1-FAT10 and FAT10 overexpression could significantly enhance the invasion and migration of HepG2 cells. Conclusion FAT10 gene overexpression in HepG2 cells can enhance the invasion and migration of liver cancer and promote the occurrence and development of liver cancer.
