目的 通过观察回转条件下培养的大鼠骨肉瘤细胞内Ⅰ型胶原的基因和蛋白表达的变化 ,探讨模拟失重对成骨细胞增殖功能的影响。 方法 对培养的大鼠骨肉瘤细胞采用回转器模拟失重 2 4、4 8、72h ,同时设 1G对照组。在各时间点提取细胞总RNA ,进行反转录PCR ,检测Ⅰ型胶原α1链 (COL Iα1)mRNA的表达情况 ,同时采用ELISA法测定细胞内Ⅰ型胶原蛋白的表达量。 结果 模拟失重 72h细胞内COL Iα1mRNA和Ⅰ型胶原蛋白的表达量均明显低于对照组。 结论 回转器模拟失重 72h使大鼠骨肉瘤细胞的增殖功能降低。
Objective To investigate the effects of simulated weightlessness on the proliferation of osteoblasts by observing the changes of type Ⅰ collagen mRNA and protein expression in osteosarcoma cells cultured in vitro. Methods The cultured rat osteosarcoma cells were simulated with a rotator to simulate the weightlessness of 2, 4, 8 and 72 hours, and 1G control group was also established. Total RNA was extracted at each time point and reverse transcription PCR was performed to detect the expression of COL Iα1 mRNA. ELISA was also used to detect the expression of collagen type Ⅰ. Results The expression of COL Ialpha mRNA and type I collagen in the 72h after simulated weightlessness were significantly lower than those in the control group. Conclusion Rotator simulated weightlessness 72h decreased proliferation of osteosarcoma cells in rats.
