目的:探讨辣椒素(Cap)对人神经母细胞瘤SH-SY5Y细胞的抑制作用、凋亡率、细胞周期、氧自由基(ROS)、细胞内钙离子(Ca2+)的影响。方法:CCK-8法检测辣椒素(50、100、150、200μmol/L)对人神经母细胞瘤SH-SY5Y细胞的抑制作用;PI染色法检测细胞周期变化;Annexin V-FITC、PI双染检测细胞凋亡;DCFHDA检测ROS水平;Fluo 3-AM检测细胞内Ca2+变化。结果:辣椒素显著抑制SH-SY5Y细胞的增殖,且呈时间和剂量依赖性。辣椒素处理12h后:G2/M期、S期减少,G0/G1期细胞增多(P<0.01),表明辣椒素将SH-SY5Y细胞限定于G0/G1期;早期凋亡率显著增加,为48.35%;辣椒素处理12h后ROS、Ca2+水平显著上调(P<0.01)。结论:辣椒素通过提高细胞内ROS、Ca2+水平,抑制人神经母细胞瘤SH-SY5Y细胞增殖,将其限定于G0/G1期,促进其凋亡,有潜力成为新型抗肿瘤天然药物。
Objective: To investigate the effects of Cap on the inhibition of human neuroblastoma SH-SY5Y cells, apoptosis rate, cell cycle, oxygen free radicals (ROS) and intracellular calcium (Ca2 +). Methods: The inhibitory effects of capsaicin (50,100,150,200μmol / L) on human neuroblastoma SH-SY5Y cells were detected by CCK-8 assay. The cell cycle was detected by PI staining. Annexin V-FITC and PI double staining Cell apoptosis was detected by flow cytometry. ROS level was detected by DCFHDA assay. Intracellular Ca2 + level was measured by Fluo 3-AM. Results: Capsaicin significantly inhibited the proliferation of SH-SY5Y cells in a time and dose-dependent manner. After treatment with capsaicin for 12h, the number of G2 / M phase and S phase decreased and the number of G0 / G1 phase cells increased (P <0.01), indicating that capsaicin limited SH-SY5Y cells to G0 / G1 phase; 48.35% respectively; ROS and Ca2 + levels were significantly increased after treatment with capsaicin for 12h (P <0.01). CONCLUSION: Capsaicin has the potential to become a new natural anti-tumor drug by increasing intracellular ROS and Ca2 + levels and inhibiting the proliferation of human neuroblastoma SH-SY5Y cells, which is limited to G0 / G1 phase and promotes apoptosis.
