目的用HPLC法测定八正胶囊中的大黄素、大黄酚。方法采用菲罗门C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-甲醇-0.1%磷酸溶液(42∶23∶35),流速1.0 mL·min-1,检测波长254 nm。结果大黄素、大黄酚的线性范围分别为0.0267~0.4272μg(r=0.9998)、0.0417~0.6677μg(r=0.9999),总大黄素、总大黄酚、游离大黄素、游离大黄酚的平均加样回收率分别为97.35%(RSD=1.7%)、98.39%(RSD=0.4%)、100.70%(RSD=1.20%)、104.41%(RSD=0.9%)。结论所用方法专属性好,准确、重复性好,可作为八正胶囊质量控制的方法之一。
Objective To determine emodin and chrysophanol in Bazheng Capsules by HPLC. Methods Phenol C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase was acetonitrile-methanol-0.1% phosphoric acid solution (42:23:35) and the flow rate was 1.0 mL · min-1. The detection wavelength was 254 nm. Results The linear ranges of emodin and chrysophanol were 0.0267-0.4272μg (r = 0.9998) and 0.0417-0.6777μg (r = 0.9999) respectively. The average additions of total emodin, total chrysophanol, free emodin and free chrysophanol were The recoveries were 97.35% (RSD = 1.7%), 98.39% (RSD = 0.4%), 100.70% (RSD = 1.20%) and 104.41% (RSD = 0.9%), respectively. Conclusion The method used has good specificity, accuracy and repeatability and can be used as one of the methods for quality control of Bazheng capsule.
