目的 研究不同浓度青蒿素衍生物双氢青蒿素、青蒿琥酯对体外培养卡氏肺孢子虫的作用。方法 接种卡氏肺孢子虫 (Pc)于人肝癌细胞系 (HepG 2 )。 4h后加入试验药物和对照试剂。双氢青蒿素药物浓度分别为 :10 0、5 0、10、5、0 5 μmol/L ;青蒿琥酯药物浓度分别为 :10 0、5 0、10、5 μmol/L ;对照药喷他脒 :15 μmol/L。以后每隔 1d取培养液观察 ,分别作六亚甲基四胺银 (GMS)和Diff Quik (DQ)染色计数Pc包囊和滋养体数目。结果 Pc滋养体和包囊在体外培养细胞上的生长高峰出现于第 5d ,双氢青蒿素浓度为 5 0 μmol/L和 10 0 μmol/L时对Pc滋养体的抑制率分别为 88 0 %与90 1% ,与喷他脒 15 μmol/L的抑制率 90 0 %相当 ;而青蒿琥酯浓度为 10 0 μmol/L时方可达到Pc滋养体抑制率 83 4%。Pc包囊抑制率 ,双氢青蒿素和青蒿琥酯浓度为 10 0 μmol/L时分别为 6 7 5 %和6 7 3% ,与喷他脒的抑制率 75 0 %之间无统计学差异。结论 双氢青蒿素和青蒿琥酯在一定浓度时对Pc的抑制作用与喷他脒相当 ;而双氢青蒿素的抑制作用略高于青蒿琥酯。两者对Pc滋养体的抑制率均高于对Pc包囊
Objective To study the effect of different concentrations of artemisinin derivatives dihydroartemisinin and artesunate on Pneumocystis carinii in vitro. Methods Pneumocystis carinii was inoculated into human hepatoma cell line HepG 2. After 4 h add the test drug and control reagent. The concentrations of dihydroartemisinin were 10 0, 50, 10, 5, 5 5 μmol / L and artesunate drug concentrations were 10 0, 50, 10, 5 and 5 μmol / L, respectively Pentamidine: 15 μmol / L. After every 1d, culture medium was taken for observation, and the numbers of Pc cysts and trophozoites were counted as hexamethylenetetramine silver (GMS) and Diff Quik (DQ) staining respectively. Results The growth peak of Pc trophozoites and cysts on cultured cells in vitro appeared on the 5th day. The inhibitory rates of Pc trophozoites at the concentrations of 50 μmol / L and 100 μmol / L of dihydroartemisinin were 88 0 % And 90 1%, respectively. The inhibition rate of Pentamidine at 15 μmol / L was 90 0%. When Artesunate concentration was 100 μmol / L, the rate of Pc nodule inhibition reached 83 4%.
