目的选择合适的大孔树脂,优化工艺参数,分离、纯化金银花叶中的黄酮。方法比较吸附率和解析率,选出合适的大孔树脂HPD100。以产品得率、黄酮得率和黄酮含量为指标,考查最佳工艺条件。结果黄酮粗品拌料与大孔树脂质量比为1∶6,装入柱径与柱长比为1∶6的层析柱中,用2倍柱体积的蒸馏水冲洗,再用70%乙醇以最大流速洗脱,收集2~8倍柱体积的洗脱液,浓缩后得到黄酮产品,得率为4.4%,含量可达91.7%。结论所选工艺操作简单、方法可靠,并能得到高纯度的产品。
Objective To select appropriate macroporous resins, optimize the process parameters, and separate and purify the flavonoids in honeysuckle leaves. Methods Adsorption rate and resolution were compared to select the appropriate macroporous resin HPD100. The yield, flavonoid yield and flavonoid content as indicators, to test the best process conditions. Results The mass ratio of the crude flavoring material to the macroporous resin was 1: 6. The column was filled with 1: 6 column diameter and column length ratio. The column was washed with 2 column volumes of distilled water and 70% The elution was carried out at a flow rate of 2 to 8 column volumes. After concentration, the flavonoid product was obtained with a yield of 4.4% and a content of 91.7%. Conclusion The selected process is simple, reliable, and can get high purity products.
