本文报告了从抗日本血吸虫单抗NP11- 4杂交瘤细胞株的基因组DNA中 ,用PCR技术扩增出抗体轻链可变区基因 ,并克隆至pUC19质粒中进行测序分析。研究表明 :该基因长 336bp ,编码 112个氨基酸 ,经结构分析及与抗体库中一般序列的比较 ,证实该序列为轻链可变区基因 ,属κ轻链SubgroupII亚类 ,由种系基因中的V与Jκ 2基因重排而成。
This paper reports the amplification of the light chain variable region gene of the antibody from the genomic DNA of the anti-Schistosoma japonicum NP11-4 hybridoma cell line by PCR and cloning into the pUC19 plasmid for sequencing analysis. The results showed that this gene was 336 bp in length and encoded 112 amino acids. The structural analysis and comparison with the general sequences in the antibody library confirmed that the gene was a light chain variable region gene and belongs to the kappa light chain Subgroup II subclass. Of V and JK2 gene rearrangement made.
