用2,5-二氟苯乙酸与茴香硫醚经傅-克反应、与3-溴-3-甲基-2-氧代丁腈成环及硝酸氧化制得2,2-二甲基-4-(2,5-二氟苯基)-5-[(4-甲磺酰基)苯基]-3(2H)-呋喃酮(7),7与乙酐反应后再经过硫酸氢钾复合盐氧化、氢氧化钠水解得4-[2,2-二甲基-3-氧代-4-(2,5-二氟苯基)-3(2H)-呋喃-5-基]苯磺酸钠(9),最后依次与磺酰氯和氨水反应制得2,2-二甲基-4-(2,5-二氟苯基)-5-[(4-氨基磺酰基)苯基]-3(2H)-呋喃酮,总收率约46%。
Using 2,5-difluorophenylacetic acid and thioanisole by Friedel-Crafts reaction and 3-bromo-3-methyl-2-oxobutyronitrile into ring and nitric acid oxidation of 2,2-dimethyl-4- (7) is reacted with acetic anhydride and then oxidized with potassium bisulfate salt , Sodium hydroxide was hydrolyzed to give sodium 4- [2,2-dimethyl-3-oxo-4- (2,5-difluorophenyl) (9) and finally with 2,2-dimethyl-4- (2,5-difluorophenyl) -5 - [(4-aminosulfonyl) phenyl] -3 (2H) -furanone in a total yield of about 46%.
