目的:评价n hsa_circ_0025853在低温减轻神经母细胞瘤细胞(SK-N-SH细胞)氧糖剥夺-复糖复氧损伤中的作用。n 方法:体外培养SK-N-SH细胞至对数生长期,采用随机数字表法分为4组(n n=20):对照组(C组)、氧糖剥夺-复糖复氧组(OGD/R组)、低温组(H组)、n hsa_circ_0025853干扰RNA(siRNA)+低温组(S+H组)。C组细胞在正常条件下培养;OGD/R组氧糖剥夺3 h后复糖复氧24 h;H组氧糖剥夺3 h后,在32 ℃低温下复糖复氧24 h。S+H组在氧糖剥夺-复糖复氧模型制备前48 h转染siRNA序列,余同H组。于复糖复氧24 h时,采用CCK-8法检测细胞活力,流式细胞术检测细胞凋亡率,qRT-PCR法检测线粒体动力相关蛋白1(Drp1) mRNA、n hsa_circ_0025853表达,Western blot法检测Drp1表达水平。n 结果:与C组比较,其余3组细胞活力降低,细胞凋亡率升高,n hsa_circ_0025853表达下调,Drp1及其mRNA表达上调(n P<0.05);与OGD/R组比较,H组和S+H组细胞活力升高,细胞凋亡率降低,n hsa_circ_0025853表达上调,Drp1及其mRNA表达下调(n P<0.05)。与H组比较,S+H组细胞活力降低,细胞凋亡率升高,n hsa_circ_0025853表达下调,Drp1及其mRNA表达上调(n P<0.05)。n 结论:低温减轻SK-N-SH细胞氧糖剥夺-复糖复氧损伤的机制与上调n hsa_circ_0025853的表达,从而降低Drp1水平有关。n “,”Objective:To evaluate the role of n hsa_circ_0025853 in hypothermia-induced reduction of oxygen-glucose deprivation and restoration (OGD/R) injury to neuroblastoma cells (SK-N-SH cells).n Methods:SK-N-SH cells were cultured n in vitro to the logarithmic phase and divided into 4 groups (n n=20 each) using a random number table method: control group (group C), group OGD/R, hypothermia group (group H) and n hsa_circ_0025853 small interfering RNA (siRNA) plus hypothermia group (group S+ H). The cells were cultured in normal culture atmosphere in group C. The cells were subjected to OGD for 3 h followed by restoration of oxygen-glucose supply for 24 h in group OGD/R.The cells were subjected to OGD for 3 h followed by restoration of oxygen-glucose supply for 24 h at 32 ℃ in group H. The cells were transfected with siRNA at 48 h before establishing OGD/R model, and the other treatments were similar to those previously described in group H. At 24 h of restoration of oxygen-glucose, cell viability was measured by Cell Counting Kit-8, apoptosis rate were measured by flow cytometry.the expression of dynamin-related protein 1 (Drp1) mRNA and n hsa_circ_0025853 was detected by quantitative real-time-polymerase chain reaction, and the expression of Drp1 protein was detected by Western blot.n Results:Compared with group C, the cell viability was significantly decreased, the apoptosis rate of cells was increased, the expression of n hsa_circ_0025853 was down-regulated, and the expression of Drp1 protein and mRNA was up-regulated in the other 3 groups (n P<0.05). Compared with group OGD/R, the cell viability was significantly increased, the apoptosis rate of cells was decreased, the expression ofn hsa_circ_0025853 was up-regulated, and the expression of Drp1 protein and mRNA was down-regulated in H and S+ H groups (n P<0.05). Compared with group H, the cell viability was significantly decreased, the apoptosis rate of cells was increased, the expression ofn hsa_circ_0025853 was down-regulated, and the expression of Drp1 protein and mRNA was up-regulated in group S+ H (n P<0.05).n Conclusion:The mechanism by which hypothermia alleviates the OGD/R injury to SK-N-SH cells is related to the up-regulation of n hsa_circ_0025853 expression, thus reducing the expression level of Drp1.n
