目的探讨矽肺患者肺泡巨噬细胞(AM)线粒体通路凋亡相关蛋白的表达及其意义。方法 52例男性矽肺患者分为矽肺壹期组18例、贰期组17例、叁期组17例,以5例男性矽尘作业工人观察对象为对照组,收集患者肺灌洗回收液,分离纯化、培养、收获AM,提取蛋白,采用免疫印迹法检测天冬氨酸特异性半胱氨酸蛋白酶(Caspase)-9、Caspase-3和细胞色素C(CytC)的蛋白相对表达水平(简称“蛋白水平”),分析其与粉尘暴露因素及矽肺发病的关系。结果 Caspase-9、CytC蛋白水平随矽肺期别增加而升高(P<0.05);矽肺壹期组、贰期组和叁期组Caspase-3蛋白水平均高于对照组(P<0.05),矽肺贰期组和叁期组蛋白水平分别高于矽肺壹期组(P<0.05)。52例矽肺患者中,开始接尘年龄≥25岁组患者Caspase-9蛋白水平高于<25岁组(P<0.05),未脱尘组CytC蛋白水平高于脱尘组(P<0.05),金矿粉尘组Caspase-9与CytC蛋白水平均高于煤矿岩尘组(P<0.05)。Caspase-9与Caspase-3蛋白水平均与矽肺期别有关联(P<0.01),CytC蛋白水平分别与矽肺期别及金矿粉尘接触有关联(P<0.01)。结论线粒体凋亡蛋白在矽肺的发生发展中可能发挥着重要的作用。
Objective To investigate the expression of mitochondrial apoptosis-related protein in alveolar macrophages (AM) of patients with silicosis and its significance. Methods Fifty-two men with silicosis were divided into two groups: silicosis group (18 cases), second stage group (17 cases) and third stage group (17 cases). Five male workers exposed to silica dust were used as the control group. Purified, cultured, harvested AM and extracted proteins. The relative expression levels of Caspase-9, Caspase-3 and CytC were detected by Western blotting (abbreviated “Protein level”), and analyzed its relationship with exposure to dust and silicosis. Results Caspase-9 and CytC protein levels increased with the increase of silicosis stage (P <0.05). The levels of Caspase-3 protein in silicosis stage I, II and III groups were significantly higher than those in control group (P <0.05) Silicosis II group and the three stage histone protein levels were higher than the silicosis group (P <0.05). Among the 52 silicosis patients, the level of Caspase-9 protein in the group of patients aged ≥25 years old was higher than that in the group <25 years old (P <0.05), while the level of CytC protein in the group without dephosphorization was higher than that in the group of de-dusting (P <0.05) The levels of Caspase-9 and CytC protein in gold dust group were higher than those in coal rock dust group (P <0.05). The protein levels of Caspase-9 and Caspase-3 were all associated with silicosis stage (P <0.01). The levels of CytC protein were correlated with stage of silicosis and gold dust contact (P <0.01). Conclusion Mitochondrial apoptosis protein may play an important role in the development of silicosis.
