目的利用DNA条形码技术对河南机场口岸截获的鼠类样本进行鉴定。方法提取该鼠类样本基因组DNA,以通用引物Bat L5310和R6036R进行细胞色素C氧化酶亚基Ⅰ(COⅠ)基因片段的扩增并测序,将测序结果与Gen Bank及BOLD Systems v3中的序列进行比对,应用Mega 4.1软件构建系统发育树。结果该鼠样本COⅠ基因扩增片段去除引物后为702 bp,序列比对结果及系统进化关系表明其与大足鼠的同源性为99.43%,该鼠样本为大足鼠。结论 DNA条形码技术在鼠类鉴定中可弥补形态学鉴定的缺陷,是一种简单有效的鼠类鉴定方法。
Objective To identify the murine samples intercepted at the port of Henan airport by DNA barcode technology. Methods The genomic DNA of the murine samples was extracted. The genomic DNA was amplified and sequenced using the universal primers Bat L5310 and R6036R. The sequencing results were compared with those of Gen Bank and BOLD Systems v3 In comparison, the phylogenetic tree was constructed using Mega 4.1 software. Results The amplified fragment of COⅠ gene of this rat sample was 702 bp after the primer was removed. The sequence alignment and phylogenetic relationship indicated that the homology was 99.43% with Daphniphyllum. Conclusion DNA barcoding technology can make up for the defects of morphological identification in the identification of rodents. It is a simple and effective rodent identification method.
