目的:研究Smad4及Smad7在不同胃组织中的表达及作用。方法:病理学证实的胃癌根治术切除的胃癌组织标本78例,胃癌旁组织78例,采用SP免疫组织化学染色技术,检测Smad4及Smad7在不同胃组织中的表达。结果:①Smad4蛋白在胃癌旁组织中的阳性表达率为69.23%,在胃癌组织中的阳性表达率为44.87%(P<0.01);随着胃癌分化程度的降低其阳性表达率降低;有淋巴结转移者阳性表达率为23.25%,无淋巴结转移者的阳性表达率为71.43%,差别亦有统计学意义(P<0.01)。②Smad7蛋白在胃癌旁组织中的阳性表达率为17.95%,在胃癌组织中的阳性表达率为89.74%(P<0.01);随着胃癌分化程度的降低其阳性表达率升高;有淋巴结转移者阳性表达率为97.67%,无淋巴结转移者的阳性表达率为80.00%,有淋巴结转移者阳性表达率高于无淋巴结转移者(P<0.05)。结论:①Smad4蛋白在胃癌组织中低表达,与癌旁组织比较差异有统计学意义(P<0.01),其表达的阳性率与胃癌的分化程度及转移有关。②Smad7蛋白在胃癌组织中高表达,与癌旁组织比较差异有统计学意义(P<0.01),其表达的阳性率与胃癌的分化程度及转移有关。
Objective: To study the expression and function of Smad4 and Smad7 in different gastric tissues. Methods: 78 cases of gastric cancer specimens resected by pathology and 78 cases of gastric cancer tissues were excised. The expression of Smad4 and Smad7 in different gastric tissues was detected by SP immunohistochemical staining. Results: ①The positive expression rate of Smad4 protein in gastric cancer tissues was 69.23%, while the positive rate in gastric cancer tissues was 44.87% (P <0.01). The positive expression rate of Smad4 protein was decreased with the differentiation degree of gastric cancer. There was lymph node metastasis The positive rate was 23.25%. The positive rate of lymph node metastasis was 71.43%. The difference was also statistically significant (P <0.01). ② The positive expression rate of Smad7 protein in gastric cancer tissues was 17.95%, while it was 89.74% (P <0.01) in gastric cancer tissues. The positive expression rate of Smad7 protein was increased with the differentiation degree of gastric cancer. The positive rate was 97.67%. The positive rate of lymph node metastasis was 80.00%. The positive rate of lymph node metastasis was higher than that without lymph node metastasis (P <0.05). Conclusion: (1) Smad4 protein is lowly expressed in gastric cancer tissues, which is significantly different from that in paracancerous tissues (P <0.01). The positive rate of Smad4 expression is correlated with the differentiation and metastasis of gastric cancer. ② Smad7 protein was highly expressed in gastric cancer tissues, which was significantly different from that in paracancerous tissues (P <0.01). The positive rate of Smad7 protein was correlated with the differentiation and metastasis of gastric cancer.
