目的:探讨健骨二仙丸提取物对大鼠体外培养成骨细胞的作用机理。方法:采用新生大鼠颅骨进行成骨细胞分离,并在含10%的胎牛血清DMEM中进行体外培养,分别加入高、低剂量的健骨二仙丸提取物并与高、低剂量Premarin组、阴性组、Tamoxifen对照组、Tamoxifen阻断对照组进行对照,用Western免疫印迹检测蛋白激酶C、抗凋亡蛋白Bag-1表达。结果:健骨二仙丸提取物能明显提高体外成骨细胞PKC的表达量和抗凋亡蛋白Bag-1的表达量。结论:健骨二仙丸提取物可促进成骨细胞增殖并明显提高PKC和抗凋亡蛋白Bag-1的表达量是其防治骨质疏松的机理之一。
Objective: To investigate the mechanism of Jiangu Erxian Wan extract on rat osteoblasts cultured in vitro. METHODS: Osteoblasts were isolated from newborn rat skull and cultured in DMEM containing 10% fetal bovine serum. High and low doses of Jiangu Erxian Wan extract were added respectively and compared with high and low dose Premarin group , Negative group, Tamoxifen control group and Tamoxifen blocking control group. Protein kinase C and anti-apoptotic protein Bag-1 were detected by Western immunoblotting. Results: Jiangu Erxian Wan extract can significantly increase the expression of PKC in vitro and the expression of anti-apoptotic protein Bag-1. Conclusion: Jiangu Erxian Wan extract can promote the proliferation of osteoblasts and significantly increase the expression of PKC and anti-apoptotic protein Bag-1, which is one of the mechanisms of prevention and treatment of osteoporosis.
