2’-去氧助间型霉素(Pentastatin,2’-deoxyco-formycin,DCF)为链丝菌属抗生素,有强烈的淋巴细胞毒性,对T和B细胞起源的恶性淋巴细胞增殖性疾患具有抗肿瘤作用。在毛细胞白血病(HCL)的DCF随机研究中其疗效优于脾切除或干扰素治疗。现综述该药的药理作用和临床疗效。一.腺苷脱氨酶(ADA)的活性分布 DCF为嘌呤代谢中的ADA抑制剂。ADA存在于肝、脾、肾等组织细胞上,但以胸腺细胞上ADA活性最高,其次为淋巴细胞。T细胞高于B细胞,浆细胞至少与T或非T非B细胞相同。急性淋巴细胞白血病(ALL)细胞上ADA活性高于正常淋巴细胞4倍,以T和非T非B ALL细胞为高。慢性淋巴细胞白血病(CLL)细胞上ADA活性低于正常淋巴细胞,但T-CLL细胞较正常T细胞高2-3倍,亦较B-CLL细胞高,化疗后随着淋巴细胞减少,ADA活性逐渐增高趋向正常。慢性粒细胞白血病急变时ADA活性增高,急淋变与急粒变之间无明显差别。T淋
