目的探讨Dickkopf-1(DKK-1)、低密度脂蛋白受体相关蛋白-5(LRP-5)和低密度脂蛋白受体相关蛋白-6(LRP-6)在卵巢上皮性癌中的表达和临床意义。方法2002年1月至2008年4月在中国医科大学附属盛京医院采用RT-PCR的方法检测56例卵巢上皮性癌、35例卵巢上皮性良性肿瘤和12例正常卵巢组织中DKK-1mRNA、LRP-5mRNA和LRP-6mRNA的表达情况,分析3种基因与各临床病理参数的关系及其在癌组织中的表达相关性。结果DKK-1mRNA,LRP-5mRNA和LRP-6mRNA在卵巢上皮性癌组织中的表达强度分别为0.90±0.32,0.71±0.27,0.80±0.28;明显高于卵巢上皮性良性肿瘤组织0.58±0.23,0.58±0.29,0.54±0.22和正常卵巢组织0.36±0.29,0.40±0.25,0.32±0.27(P均<0.0001)。DKK-1mRNA、LRP-5mRNA和LRP-6mRNA表达强度与临床分期(P=0.003~0.020)和病理分级(P=0.000~0.028)密切相关。DKK-1mRNA与LRP-5mRNA和LRP-6mRNA在卵巢上皮性癌中的表达强度存在统计学相关性(r=0.341,P<0.0001;r=0.815,P<0.0001)。结论DKK-1、LRP-5和LRP-6mRNA在卵巢上皮性癌中表达增强,其表达强度存在相关联系。DKK-1、LRP-5和LRP-6在卵巢上皮性癌的发生、发展过程中起重要作用。
Objective To investigate the expression of Dickkopf-1 (DKK-1), low density lipoprotein receptor associated protein-5 (LRP-5) and low density lipoprotein receptor related protein- 6 (LRP-6) in epithelial ovarian cancer And clinical significance. Methods From January 2002 to April 2008, DKK-1 mRNA in 56 cases of epithelial ovarian cancer, 35 cases of benign epithelial ovarian tumor and 12 cases of normal ovarian tissue were detected by RT-PCR in Shengjing Hospital affiliated to China Medical University. LRP-5mRNA and LRP-6mRNA, and to analyze the relationship between the three genes and clinicopathological parameters and their expression in cancer tissues. Results The expressions of DKK-1 mRNA, LRP-5 mRNA and LRP-6 mRNA in ovarian epithelial carcinoma were 0.90 ± 0.32, 0.71 ± 0.27 and 0.80 ± 0.28, respectively, which were significantly higher than those in ovarian epithelial benign tumor (0.58 ± 0.23, 0.58 ± 0.29, 0.54 ± 0.22 and normal ovarian tissue 0.36 ± 0.29, 0.40 ± 0.25, 0.32 ± 0.27 (P <0.0001, respectively). The expressions of DKK-1mRNA, LRP-5mRNA and LRP-6mRNA were closely related to clinical stage (P = 0.003-0.020) and pathological grade (P = 0.000-0.028) There was a statistically significant correlation between the expression intensity of DKK-1 mRNA and LRP-5 mRNA and LRP-6 mRNA in epithelial ovarian cancer (r = 0.341, P <0.0001; r = 0.815, P <0.0001). Conclusion The expression of DKK-1, LRP-5 and LRP-6 mRNA in epithelial ovarian cancer is increased, and the expression intensity of DKK-1, LRP-5 and LRP-6 are correlated. DKK-1, LRP-5 and LRP-6 play an important role in the development and progression of ovarian epithelial carcinoma.
