目的 :研究经典方剂十全大补汤(以下简称sqdbt)联合化疗药物5-氟尿嘧啶(以下简称5-fu)对HT29移植性大肠癌小鼠的抑瘤作用及其作用机制。方法 :采用HT29移植性大肠癌瘤株制作大肠癌小鼠模型。24 h后随机分成单纯肿瘤组、sqdbt组、5-fu组、sqdbt+5-fu组,给药4周后,计算抑瘤率,对肿瘤组织进行病理切片HE染色观察及免疫组化方法检测γ-IFN值。结果 :经药物治疗后,HT29大肠癌小鼠体内肿瘤生长受到不同程度的抑制,sqdbt+5-fu组抑瘤率最高;病理切片可见治疗组肿瘤细胞数目、形态、大小均好于对照组;免疫组化检测γ-IFN值sqdbt+5-fu组最高。结论 :十全大补汤具有提高HT29大肠癌小鼠生存质量的作用,并可以抑制HT29大肠癌小鼠肿瘤生长、提高机体免疫力、对化疗药物治疗肿瘤有减毒增效的作用。
Objective: To study the anti-tumor effect and its mechanism of the classic prescription of Shiquan Dabu Tang (hereinafter referred to as sqdbt) combined with 5-fluorouracil (5-fu) chemotherapy on HT29 transplanted colorectal cancer in mice. Methods: The colorectal cancer mouse model was made by HT29 colon carcinoma cell line. Twenty-four hours later, the mice were randomly divided into simple tumor group, sqdbt group, 5-fu group, and sqdbt + 5-fu group. After 4 weeks of administration, the tumor inhibition rate was calculated and the tumor tissues were examined by HE staining and immunohistochemistry γ-IFN value. Results: The tumor growth of HT29 colorectal cancer mice was inhibited to some extent after drug treatment, and the inhibition rate was the highest in the sqdbt + 5-fu group. The number, shape and size of the tumor cells in the treatment group were better than those in the control group; Immunohistochemical detection of γ-IFN was the highest in the sqdbt + 5-fu group. Conclusion: Shiquan Dabu Tang can improve the quality of life in HT29 colorectal cancer mice, and can inhibit tumor growth in HT29 colorectal cancer mice, improve immunity and reduce the toxicity and toxicity of chemotherapy drugs.
