目的:研究雷公藤多苷(tripterginum wilfordii polyglycoside,TWP)配伍黄芪总皂苷(astragalosides,ASTs)对佐剂性关节炎(AA)大鼠的治疗效果及肝肾功能的影响。方法:弗式完全佐剂建立AA大鼠模型,造模后第1d各组给药,连续28d,测量大鼠原发性和继发性足肿胀程度,检测血清中白介素-1β(IL-1β)和丙二醛(MDA)水平,检测大鼠的肝肾功能。结果:(1)相较于AA模型组,雷公藤多苷组(雷公藤多苷10mgkg-1,1次/d)、黄芪总皂苷组(黄芪总皂苷100mgkg-1,1次/d)和配伍组(雷公藤多苷10mgkg-1+黄芪总皂苷100mgkg-1,1次/d)中大鼠足爪肿胀程度均显著下降(P<0.01),其中以配伍组大鼠关节炎症状改善最为明显;(2)模型组大鼠血清IL-1β水平明显升高,而经过治疗后血清中IL-1β含量下降,且以配伍组降低最为显著;(3)与模型组相比,雷公藤多苷组大鼠的谷草转氨酶(AST)、谷丙转氨酶(ALT)、丙二醛(MDA)、血尿素氮(BUN)和肌酐(Cr)均显著增高(P<0.01或P<0.05);(4)与雷公藤多苷组相比,配伍组大鼠的AST、ALT、MDA、BUN和Cr均显著降低。结论:雷公藤多苷配伍黄芪总皂苷使用可显著降低血清中炎症因子水平,减轻AA大鼠的关节炎症状,并具有减毒增效的作用。
Objective: To study the effects of tripterginum wilfordii polyglycoside (TWP) combined with astragalosides (ASTs) on the therapeutic effect and liver and kidney function of adjuvant arthritis (AA) rats. Methods: AA model rats were established by Freund’s complete adjuvant, and the rats in each group were administrated on the 1st day after modeling for 28 days. The extent of primary and secondary paw swelling in rats was measured. The level of interleukin-1β (IL-1β ) And malondialdehyde (MDA) levels were detected in rats liver and kidney function. Results: (1) Compared with the AA model group, tripterygium glycosides group (Tripterygium glycosides 10mg kg-1,1 times / d), Astragalus total saponins group (Astragalus total saponins 100mg kg-1 / d) and compatibility group (Tripterygium glycosides 10mgkg-1 + Astragalus total saponin 100mgkg-1,1 times / d) were significantly decreased (P <0.01), which compatibility (2) The level of IL-1β in the model group was significantly increased, but the level of IL-1β in the serum of the model group decreased after treatment, and the most significant decrease was found in the compatibility group; (3) Compared with model group, the contents of AST, ALT, MDA, BUN and Cr were significantly increased in tripterygium glycosides group (P < 0.01 or P <0.05). (4) Compared with tripterygium glycosides group, the levels of AST, ALT, MDA, BUN and Cr in the compatibility group were significantly decreased. Conclusion: The combination of Tripterygium glycosides and Astragalus total saponin can significantly reduce the levels of inflammatory cytokines in serum, alleviate the arthritis symptoms in AA rats and have the effect of attenuating toxicity and increasing synergism.
