目的:采用分光光度法研究了脯氨酸与2,3-二氯-5,6-二氰-1,4-对苯醌(DDQ)的荷移反应。方法:在常温下,脯氨酸与DDQ在十二烷基硫酸钠溶液及pH=10.0的三酸缓冲溶液中,反应10 min,可生成稳定的荷移络合物。结果:荷移络合物的组成比为1∶1,最大吸收波长λmax=523nm,表观摩尔吸光系数ε=1.60×103L.mol-1.cm-1,在10μg ml-1~70μg ml-1的范围内符合比尔定律,回收率在97.7%~100.5%的范围内,桑德尔灵敏度S=7.2×10-2μg cm-2,相对标准偏差不大于1.5%。结论:应用本方法测定脯氨酸样品的含量,结果与文献方法一致。
OBJECTIVE: To study the charge-transfer reaction of proline with 2,3-dichloro-5,6-dicyano-1,4-p-benzoquinone (DDQ) by spectrophotometry. Methods: Under normal temperature, Proline and DDQ were reacted in sodium lauryl sulfate solution and trisacidic buffer solution with pH = 10.0 for 10 min to produce stable charge-transfer complexes. Results: The composition ratio of charge-transfer complex was 1: 1, the maximum absorption wavelength λmax was 523nm, the apparent molar absorptivity ε was 1.60 × 103L.mol-1.cm-1, Beer’s law was obeyed in the range of 1 and the recoveries ranged from 97.7% to 100.5%. The sensitivity of SANDAL was 7.2 × 10-2μg cm-2 and the relative standard deviation was less than 1.5%. Conclusion: The content of proline in this method was determined. The result was consistent with the literature method.
