目的探讨益气活血化痰法对大鼠酒精性肝纤维化的影响及机制。方法将120只SD大鼠随机分成对照组、模型组、秋水仙碱组、小剂量中药组、中剂量中药组、大剂量中药组各20只。除对照组外,其余各组大鼠均用56°酒精灌胃[15ml/(kg.d)-1]致肝纤维化。同时,各小、中、大剂量中药组分别以10g/(kg.d)-1、20g/(kg.d)-1、40g/(kg.d)-1益气活血化痰法中药组方灌胃,秋水仙碱组以秋水仙碱灌胃[2mg/(kg.d)-1],模型组予等量生理盐水灌胃。6个月后处死取肝,采用HE染色考察各组大鼠肝脏的纤维化程度,免疫组化法检测肝组织转化生长因子-β1(TGF-β1)蛋白表达,用RT-PCR法检测各组大鼠肝组织中TGFβ-1和Smads3、Smads4、Smads7的核酸水平表达。结果不同剂量中药组及秋水仙碱组的纤维化分期均与模型组比较差异有统计学意义(P<0.05或P<0.01);与模型组比较,各中药组及秋水仙碱组的TGFβ-1蛋白表达均显著降低(P<0.01),且大剂量中药组较秋水仙碱组TGFβ-1蛋白显著降低(P<0.01);不同剂量与治疗后TGF-β1、Smad3、Smad4、Smad7各核酸分子的表达水平均有相关性。结论益气活血化痰法能下调酒精性肝纤维化大鼠TGFβ-1、Smad3、Smad4 mRNA表达水平并上调smad7 mRNA表达水平,干预了Smads信号通路的表达。
Objective To investigate the effect and mechanism of Yiqi Huoxue Huatan method on alcoholic liver fibrosis in rats. Methods 120 SD rats were randomly divided into control group, model group, colchicine group, small dose of traditional Chinese medicine group, medium dose of traditional Chinese medicine group and high dose of traditional Chinese medicine group of 20 rats. Conclusion Yiqi Huoxue Sputum Decoction can down-regulate the expression of TGFβ-1, Smad3 and Smad4 mRNA and up-regulate the expression of smad7 mRNA in alcoholic liver fibrosis rats and interfere the expression of Smads signaling pathway.
