目的探讨托吡酯(TPM)对癫大鼠血清瘦素水平和下丘脑弓状核神经肽Y(NPY)表达的影响及两者与体质量变化的关系。方法115只大鼠随机分成分别为空白组、TPM对照组、模型组和TPM治疗组。称重1次/3d,分别于2、4、6周末用放射免疫法测血清瘦素水平,用免疫组织化学染色法测下丘脑弓状核NPY表达。结果TPM对照组和TPM治疗组实验第4周出现体质量下降(P<0.05),同时在第2、4周末两组血清瘦素较空白组、模型组明显升高(P<0.05),且随时间下降。TPM治疗组与TPM对照组大鼠NPY神经元表达较空白组和模型组升高(P<0.05),且随时间渐上升。结论TPM能提高大鼠血清瘦素水平,从而降低体质量,但瘦素升高使体质量下降与NPY无关。
Objective To investigate the effect of topiramate (TPM) on the serum leptin levels and the neuropeptide Y (NPY) expression in the hypothalamus of epileptic rats and the relationship between them and the changes of body weight. Methods A total of 115 rats were randomly divided into blank control group, TPM control group, model group and TPM treatment group. The levels of serum leptin were measured by radioimmunoassay at the end of 2, 4 and 6 weeks respectively. The expression of NPY in the arcuate nucleus of the thalamus was measured by immunohistochemical staining. Results The body mass of TPM control group and TPM treatment group decreased at 4 weeks (P <0.05), and at the end of the 2nd and the 4th week, the level of serum leptin in the two groups was significantly higher than that in the blank group and the model group (P <0.05) Fall with time. Compared with blank group and model group, the expression of NPY neurons in TPM treatment group and TPM control group increased (P <0.05), and gradually increased with time. Conclusion TPM can increase the level of serum leptin in rats, thereby reducing the body weight, but the increase of leptin body mass decline has nothing to do with NPY.
