研究微波预处理,热水浸提提取香椿叶总多糖的最佳工艺条件。以香椿叶总多糖提取量为考察指标,单因素考察了润湿时间、润湿料液比、微波功率、微波处理时间对香椿叶总多糖提取效果的影响。在单因素实验的基础上,用正交实验法对香椿叶总多糖的微波预处理工艺条件进行优化,选用L9(34)进行正交实验,通过实验设计优选出香椿叶总多糖微波预处理溶剂回流提取的最佳工艺条件并验证、确定提取次数。结果表明:在香椿叶质量10.0g,提取温度100℃,提取时间2h,浸提料液比1:15条件下,微波辅助处理香椿叶的最优工艺条件为:润湿料液比1:5,润湿时间1.5h,微波处理功率1000W,微波处理时间60s。提取次数5次。在最佳参数组合下,提取5次每克香椿叶可提取总多糖为46.156mg。
The optimum conditions for extracting total polysaccharide from Toona sinensis leaves by microwave pretreatment and hot water extraction were studied. Taking the total polysaccharide of Toona sinensis leaves as index, single factor was used to study the effects of wetting time, liquid to solid ratio, microwave power and microwave treatment time on the extraction of total polysaccharide from toona sinensis leaves. On the basis of single factor experiment, orthogonal experiment was used to optimize the microwave pretreatment of Toona sinensis total polysaccharide. Orthogonal test was carried out with L9 (34), and the total polysaccharide microwave pretreatment solvent Reflux extracted the best process conditions and verification, to determine the number of extraction. The results showed that the optimum conditions for microwave assisted processing of Toona sinensis leaves were as follows: the mass ratio of wet to liquid 1: 5 , Wetting time 1.5h, microwave processing power 1000W, microwave processing time 60s. Extract 5 times. Under optimal parameters, total polysaccharides extracted per gram of Toona sinensis leaves was 46.156 mg.
