本文介绍反相高压液相层析法测定血清中的甲氨蝶呤及7-羟基甲氨蝶呤。在浓度为10~(-7)M范围内,本法对甲氨蝶呤和7-羟基甲氨蝶呤的回收率分别为90%和70%以上。实验表明,次要代谢产物2,4-二氨基-10-甲基蝶酸由于保留时间长(28.1分),甲酰四氢叶酸则由于保留时间短(4.3分),对本法均无影响。以N-{4-[(2,4-二氨基-6-喹唑啉)甲氨基]苄基}天冬氨酸为内标。在给病人输注3500~5000mg/m~2高剂量甲氨蝶
This article describes the determination of methotrexate and 7-hydroxy methotrexate in serum by reversed-phase high pressure liquid chromatography. The recoveries of methotrexate and 7-hydroxymetaphotene were 90% and 70% respectively in the range of 10 ~ (-7) M. Experiments showed that the secondary metabolites 2,4-diamino-10-methyl-pheneic acid due to a long retention time (28.1 points), leucovorin due to the short retention time (4.3 points), the law had no effect.
