采用反相高效液相色谱柱前衍生法测定兔脑匀浆液中氨基酸的含量。衍生试剂是异硫氰酸苯酯(PITC)。17种氨基酸衍生物在UctrosphereC18柱上经0.05mol/L醋酸铵(pH6.67)和乙腈的梯度淋洗而分离。包括兴奋性氨基酸递质Asp,Glu,GABA在内的8种氨基酸在20~308mmol/L的范围内线性关系良好。保留时间和峰面积的变异系数分别小于2%和0.5%.方法准确,重现性好。定量测定了兔脑中含量较高的上述8种氨基酸
The content of amino acids in homogenate of rabbit brain was determined by RP-HPLC. The derivatizing agent is phenyl isothiocyanate (PITC). Seventeen amino acid derivatives were separated on a Uctrosphere C18 column by a gradient of 0.05 mol / L ammonium acetate (pH 6.67) and acetonitrile. The eight kinds of amino acids including excitatory amino acid transmitters Asp, Glu and GABA had good linearity in the range of 20 ~ 308mmol / L. The coefficient of variation of retention time and peak area were less than 2% and 0.5%, respectively. The method is accurate and reproducible.
