目的探讨全反式维A酸(ATRA)对雌孕激素诱导性子宫肌瘤大鼠模型子宫肌瘤发生、发展的影响。方法选取SPF级雌性Wistar大鼠60只,编号后采用随机数字表法分为空白组、模型组和ATRA组,每组20只。采用苯甲酸雌二醇对模型组和ATRA组进行肌肉注射[0.5 mg(kg·d),12周]、黄体酮肌注[5.9 mg(kg·d),12周],ATRA组于第10周起给予ATRA肌注(0.5 mg/d,2周)。分析比较各组大鼠子宫重量、子宫系数等指标,以及血清激素、维A酸受体α(RARα)、一氧化氮合酶(i NOS)的水平。结果模型组、ATRA组大鼠的子宫重量、子宫系数、左右宫角长度值、宫角最大值均显著高于空白组(P<0.05),ATRA组大鼠的子宫重量、子宫系数、左右宫角长度值、宫角最大值、宫颈长度值均显著小于模型组(P<0.05)。ATRA组大鼠的雌二醇(E2)、孕酮(P)、促黄体生成素(LH)值均显著小于模型组(P<0.05)。模型组、ATRA组大鼠的子宫组织RARɑ表达显著高于空白组(P<0.05),模型组大鼠i NOS表达显著高于空白组、ATRA组(P<0.05),ATRA组的子宫组织中i NOS表达显著高于空白组(P<0.05)。结论 ATRA可通过与RARɑ结合抑制NO的增加,调节激素水平,从而达到抑制子宫平滑肌细胞增殖的目的。
Objective To investigate the effect of all-trans retinoic acid (ATRA) on the occurrence and development of uterine fibroids in estrogen-progestogen-induced uterine fibroids in rats. Methods Sixty SPF female Wistar rats were selected and divided into blank group, model group and ATRA group after random number table. The rats in model group and ATRA group were intramuscularly injected with estradiol benzoate [0.5 mg (kg · d) for 12 weeks], intramuscular injection of progesterone [5.9 mg (kg · d) for 12 weeks) ATRA intramuscular injection (0.5 mg / d for 2 weeks). The levels of uterine weight, uterine coefficient and serum hormone, RARα, and nitric oxide synthase (iNOS) in rats were analyzed. Results The weight of uterus, the coefficient of uterus, the length of left and right temples and the maximum of cornual angle in model group and ATRA group were significantly higher than those in blank group (P <0.05). The weight of uterus, uterine coefficient, The length of the angle, the maximum angle of the uterus, and the length of the cervix were significantly less than those of the model group (P <0.05). The estradiol (E2), progesterone (P) and luteinizing hormone (LH) in the ATRA group were significantly lower than those in the model group (P <0.05). The expression of RARɑ in model group and ATRA group was significantly higher than that in blank group (P <0.05). The iNOS expression in model group was significantly higher than that in blank group and ATRA group (P <0.05) i NOS expression was significantly higher than the blank group (P <0.05). Conclusion ATRA can inhibit the proliferation of uterine smooth muscle cells by inhibiting the increase of NO and regulating the hormone level by combining with RARɑ.
