目的:建立芦笋中门冬氨酸的含量测定方法。方法:采用柱前衍生RP-HPLC法,分析色谱柱为氨基酸分析专用ODS柱(4.6 mm×250 mm,5μm),柱温27℃,梯度洗脱,检测波长为360 nm。结果:门冬氨酸与其它氨基酸组分基线分离良好,在所试浓度(2.01~50.24)μg.mL-1范围内与峰面积值呈良好的线性关系,回归方程为Y=7.5×103X-1.3×104,r2=0.999 9(n=5);平均回收率为98.28%,RSD为1.85%(n=6)。结论:该法样品处理简单,准确性高,精密度好,可用于芦笋中门冬氨酸的含量测定。
Objective: To establish a method for determination of aspartic acid in asparagus. METHODS: Precolumn derivatization RP-HPLC method was used to analyze the column for amino acid analysis ODS column (4.6 mm × 250 mm, 5μm), the column temperature 27 °C, gradient elution, the detection wavelength was 360 nm. Results: The baseline separation of aspartic acid and other amino acid components was good. The linear relationship between the aspartic acid and other amino acid components was in a good linear relationship with the value of the peak area (2.01~50.24) μg.mL-1. The regression equation was Y=7.5×103X- 1.3×104, r2=0.999 9 (n=5); the average recovery rate was 98.28% and the RSD was 1.85% (n=6). Conclusion: The method is simple, accurate and accurate. It can be used for the determination of aspartic acid in asparagus.
