目的:探讨组蛋白脱乙酰化酶抑制剂苯丁酸钠(PB)体外对Kasumi-1细胞的生长抑制作用。方法:应用MTT比色法、锥虫蓝拒染法和集落细胞培养连续传代方法观察PB对细胞的生长抑制作用和自我更新能力的影响。结果:PB呈剂量依赖性抑制Kasumi-1细胞的生长,半数抑制浓度(IC50)约为2.3 mmol/L。结论:脱乙酰化酶抑制剂苯丁酸钠抑制Kasumi-1细胞的生长。
Objective: To investigate the inhibitory effect of sodium butyrate (PB), a histone deacetylase inhibitor, on the growth of Kasumi-1 cells in vitro. Methods: MTT colorimetric assay, trypan blue exclusion assay and colony-forming cell culture method were used to observe the effects of PB on the growth inhibition and self-renewal of cells. RESULTS: PB inhibited the growth of Kasumi-1 cells in a dose-dependent manner with an IC50 of about 2.3 mmol / L. Conclusion: Sodium butyrate, a deacetylase inhibitor, inhibits the growth of Kasumi-1 cells.
