目的 研究抗病毒抗生素 17997对单纯疱疹病毒 1型 (HSV - 1)DNA及蛋白质合成的影响。方法 应用氯化铯密度梯度离心 ,PAGE电泳及WesternBlot方法分别研究HSV - 1DNA及蛋白质合成。结果 179970 5 μmol/L ,1 0 μmol/L ,2 0 μmol/L及 4 0 μmol/L分别抑制HSV - 1DNA合成6 3 1% ,74 1% ,93 9%及 10 0 %。 179974 0 μmol/L对细胞DNA合成无影响。同上 4种浓度 17997对VERO细胞蛋白质合成无影响 ,但可选择性地抑制病毒蛋白质合成。结论 17997对细胞无毒浓度可选择性地抑制HSV - 1DNA及蛋白质合成
Objective To study the effect of antiviral antibiotic 17997 on the herpes simplex virus type 1 (HSV - 1) DNA and protein synthesis. Methods HSV - 1 DNA and protein synthesis were studied by cesium chloride density gradient centrifugation, PAGE electrophoresis and Western blotting. Results 179970 5 μmol / L, 10 μmol / L, 20 μmol / L and 40 μmol / L inhibited the synthesis of HSV - 1 DNA by 61.3%, 74.1%, 93.9% and 100% respectively. 179974 0 μmol / L has no effect on cell DNA synthesis. The above four concentrations of 17997 have no effect on the protein synthesis of VERO cells, but selectively inhibit viral protein synthesis. Conclusion 17997 can selectively inhibit HSV - 1 DNA and protein synthesis in nontoxic cells
