目的 :分析不明原因感音神经性聋 (ISNHL )患者血浆中热休克蛋白 (HSPs)抗体的成分 ,探讨 HSPs抗体存在的意义。方法 :以纯化的人类重组 H SP2 7、HSP6 0、HSP71、HSP90 α和 HSP90 β为抗原 ,用免疫转印迹技术检测 ISNHL、系统性红斑狼疮及正常健康者血浆中相应的抗体。结果 :ISNH L 组血浆中 H SP2 7、HSP6 0、H SP71、HSP90 α和 HSP90 β抗体阳性率分别为 12 .1%、7.3%、5 1.2 %、4.8%、0 % ,其中 HSP71抗体阳性率显著高于系统性红斑狼疮组和正常对照组 (P <0 .0 1)。结论 :HSP71抗体是 ISNHL 患者体内主要的热休克蛋白抗体 ,该抗体的存在预示某些 ISNHL 可能与自身免疫有关。
OBJECTIVE: To analyze the composition of plasma heat shock protein (HSPs) antibodies in patients with sensorineural deafness of unknown origin (ISNHL) and to explore the significance of the presence of HSPs antibodies. Methods: The purified human recombinant Hsp7, HSP60, HSP71, HSP90 α and HSP90 β were used as the antigen in the plasma of ISNHL, systemic lupus erythematosus and normal controls by immunoblotting. Results: The positive rates of plasma Hsp27, HSP60, Hsp71, HSP90 α and HSP90 β in ISNH L group were 12.1%, 7.3%, 5.2%, 4.8% and 0% Which was significantly higher than that in systemic lupus erythematosus group and normal control group (P <0.01). Conclusion: The HSP71 antibody is the major heat shock protein in ISNHL patients. The presence of this antibody indicates that some ISNHL may be involved in autoimmunity.
