目的:观察补肾复方逆转压力负荷增加大鼠心肌纤维化的作用。方法:采用腹主动脉狭窄所致压力负荷增加大鼠左室重构模型,将雄性SD大鼠60只随机分为假手术组、模型组、卡托普利组、补肾复方大剂量组、补肾复方小剂量组,观察用药4周后左室心肌病理形态胶原染色、左室心肌间质超微结构、左室心肌Ⅰ型胶原免疫组织化学染色、左室心肌Ⅲ型胶原免疫组织化学染色等指标的改变。结果:模型组Ⅰ型胶原的平均光密度与假手术组比显著升高(P<0.01),卡托普利组、补肾复方大剂量组、小剂量组的Ⅰ型胶原平均光密度均较模型组显著下降(P<0.01)。模型组Ⅲ型胶原的平均光密度与假手术组比显著升高(P<0.01),卡托普利组、补肾复方大剂量组、小剂量组的Ⅲ型胶原平均光密度均较模型组显著下降(P<0.01)。左室心肌病理形态胶原染色、左室心肌间质超微结构的改变与Ⅰ型胶原和Ⅲ型胶原的改变基本一致。结论:补肾复方具有与卡托普利基本一致的逆转心肌纤维化的作用。
Objective: To observe the effect of Bushen Recipe in reversing pressure overload on myocardial fibrosis in rats. Methods: Left ventricular remodeling was induced by pressure overload in abdominal aorta. Sixty male Sprague-Dawley rats were randomly divided into sham operation group, model group, captopril group, high-dose Bushen compound group, In the compound low-dose group, the left ventricular myocardial collagen morphology, the ultrastructure of left ventricular myocardium, the type Ⅰ collagen immunohistochemical staining of left ventricular myocardium and the expression of type Ⅲ collagen in left ventricular myocardium were observed 4 weeks after treatment Change. Results: The mean optical density of type Ⅰ collagen in model group was significantly higher than that in sham operation group (P <0.01). The average optical density of type Ⅰ collagen in captopril group, Group decreased significantly (P <0.01). The mean optical density of type Ⅲ collagen in model group was significantly higher than that in sham operation group (P <0.01). The average optical density of type Ⅲ collagen in captopril group, high-dose tonifying kidney capsule group and low-dose group were significantly higher than those in model group Decreased (P <0.01). Left ventricular myocardial morphological collagen staining, left ventricular myocardial ultrastructure changes and type I collagen and type Ⅲ collagen changes are basically the same. Conclusion: Bushen compound has the same effect with captopril in reversing myocardial fibrosis.
